Micropropagation of the avocado 'Ouro Verde' through culture of nodal segments

The present work was carried out in order to solve problems about contamination, oxidation and propagation in in vitro cultures of avocado 'Ouro Verde' nodal segments. The surface plant was sterilized with ethanol (70%), for 1 min followed by sodium hypochlorite (1%) for 10 min. The axillary buds growth was induced in growth medium supplemented by 3 mg/L of BAR The explants oxidation was avoided by inorganic salts reduction at one-half strength in growth medium. The rate of 45% of rooting shoots occurred after three days of growth in a medium containing 25 mg/L of AIB and subsequently change to an auxin-free medium with salts diluted and 1 mg/L of activated charcoal.
