Micropropagation of 'Valência' orange through culture of axillary buds in vitro

Abstract

Plants of 'Valência' (Citrus sinensis Osb.) were obtained by vegetative propagation through in vitro culture of nucellar embryos from seeds of developing fruits, twelve weeks after pollination. The axillary buds were excised and cultured on "MS" media supplemented with all of the possible combinations of NAA and BAP at 0.0, 0.2, 2.0 and 5.0 mg/l concentrations. The highest frequency of bud multiplication was observed on NAA-0.2 + BAP-0.2 mg/l with 6.18 new buds for each explant. The shoots were removed for "MS" media additioned by NAA-0.1, 0.5, 1.0, 5.0, IBA-1.0, 2.0, 5.0, 10.0, NAA-0.1, 1.0 or 5.0 + IBA-2.0, NAA-1.0 + IBA-5.0, NAA-5.0 + BAP-0.1 and GA3 - 5.0 mg/i. The best results (90%) were showed by NAA-0.1 and 1.0 + IBA-2.0 mg/l.
